Efficient purification of cell culture-derived classical swine fever virus by ultrafiltration and size-exclusion chromatography

摘要: Large-scale production of cell culture-based classical swine fever virus (CSFV) vaccine is hampered by the adverse reactions caused by contaminants from host cell and culture medium. Hence, we have developed an efficient method for purifying CSFV from cell-culture medium. Pure viral particles were obtained with two steps of tangential-flow filtration (TFF) and size-exclusion chromatography (SEC), and were compared with particles from ultracentrifugation by transmission electron microscopy (TEM), infectivity and recovery test, and real time fluorescent quantitative PCR (FQ-PCR). TFF concentrated the virus particles effectively with a retention rate of 98.5%, and 86.2% of viral particles were obtained from the ultrafiltration retentate through a Sepharose 4 F F column on a biological liquid chromatography system. CSFV purified by TFF-SEC or ultracentrifugation were both biologically active from 1.0×10 TCID ·mL to 3.0×10 TCID ·mL , but the combination of TFF and SEC produced more pure virus particles than by ultracentrifugation alone. In addition, pure CSFV particles with the expected diameter of 40–60 nm were roughly spherical without any visible contamination. Mice immunized with CSFV purified by TFF-SEC produced higher antibody levels compared with immunization with ultracentrifugation-purified CSFV ( <0.05). The purification procedures in this study are reliable technically and feasible for purification of large volumes of viruses.

Characterization of particle deposition during crossflow filtration as influenced by permeate flux andcrossflow velocity using a microfluidic filtration system

摘要: Particle deposition during crossflow filtration is significantly influenced by the operating conditions, in particular the permeate flux and crossflow velocity. However, there is a lack of detailed knowledge about how deposit layer structures and distributions depend on operating parameters. This study uses a microfluidic visualisation filtration system to examine the influence of operating conditions on the deposition process during crossflow ultrafiltration from a microscopic perspective. Increasing the permeate flux caused an increasing amount of deposition and a thicker deposit layer. Higher crossflow velocities reduced the extent of deposition. The degree of deposition varied over a range of operating conditions due to the altered hydrodynamic forces exerted on the particles, which can be examined by the deposition probability according to an existing model. Building on this, an empirical correlation between the deposition probability and volume of deposition as function of filtration time was developed, which gave good agreement with experimental results. The effect of solution conditions was also involved in this correlation as a interaction energies. This could be useful for predicting the dynamic deposition process during crossflow filtration over a range of operating and solution conditions.

本文研究了H2O、酚醛树脂胶黏剂(PF)、脲醛树脂胶黏剂(UF)和改性大豆蛋白胶(MSA)在毛竹材不同立杆高度弦切面的润湿性能,通过测定这些润湿液体在竹材弦切面上的接触角,建立润湿液体在毛竹材弦切面的动态润湿模型,分析毛竹材的不同高度对润湿液体动态润湿性能的影响,并对比分析各种润湿液体在毛竹材弦切面的润湿性能差异。

Membrane fouling control by ultrasound in an anaerobic membrane bioreactor

摘要: In this study, ultrasound was used to control the membrane fouling online in an anaerobic membrane bioreactor (AMBR). Short-term running experiments were carried out under different operating conditions to explore feasible ultrasonic parameters. The experimental results indicated that when the crossflow velocity was more than 1.0 m/s, membrane fouling could be controlled effectively only by hydrodynamic methods without ultrasound. When ultrasound was applied, an ultrasonic power range of 60–150 W was suitable for the membrane fouling control in the experimental system. The experimental results showed that the membrane fouling was controlled so well that membrane filtration resistance (Σ) could stay at 5 × 10 m for more than a week with the crossflow velocity of 0.75 m/s, which equaled the effect of crossflow velocity of more than 1.0 m/s without ultrasound.

树盘干缩异向性引起应变的测算及分析

摘要:

研究探讨了干燥过程中树盘受抑制干缩应变、自由干缩应变、弹性应变、黏弹性蠕变应变以及机械吸附蠕变的图像解析测算法;运用该方法测算了白桦树盘常规缓慢干燥(含水率分布均匀)过程中干缩异向性引起的弦向各应变,分析了干燥过程中不同含水率阶段的应力状态及应力与各应变的关系。结果表明:应变的图像解析测算法可满足精度要求;树盘干燥至fiber saturation point( FSP)以下,弦向首先受拉伸应力作用,随着干燥的进行,拉伸应力转变为压缩应力;应力方向与各应变对应关系不同,与黏弹性蠕变应变无明显对应关系,与机械吸附蠕变基本对应。

Polyhedral linkages obtained as assemblies of planar link groups

The study aims to devise means of obtaining polyhedral linkages for homothetic deployment of polyhedral shapes by embedding planar link groups in faces of the polyhedral shape of interest. The questions of which polyhedral shapes may be suitable for such a purpose and what are the compatibility conditions for spatially assembling planar link groups are addressed. Homohedral and tangential polyhedral shapes are found to be suitable for the task and some examples of linkages are worked out.

various types of 3-hydroxy-3-methylglutaryl coenzyme A reductase inhibitors on changes in glomerular filtration

摘要: Few long-term follow-up studies have compared the changes in renal function according to the type of statin used in Korea. We compared the long-term effects of statin intensity and type on the changes in the glomerular filtration rate (GFR). We extracted data of patients who took statin for the first time. We analyzed whether or not different statins affect the changes in GFR at 3 months after baseline and 4 years after. We included 3678 patients and analyzed the changes in GFR. The GFR decreased by 3.2%±0.4% on average 4 years after the first statin prescription, indicating statistically significant deterioration (from 83.5±0.4 mL/min/1.73 m to 79.9±0.4 mL/min/1.73 m , <0.001). When comparing the GFR among different statins, significant differences were observed between atorvastatin and fluvastatin (−5.3%±0.7% vs. 1.2%±2.2%, <0.05) and between atorvastatin and simvastatin (−5.3%±0.7% vs. −0.7%±0.8%, <0.05). In pitavastatin (odds ratio [OR]=0.64, 95% confidence interval [CI]=0.46–0.87, <0.005) and simvastatin (OR=0.69, 95% CI=0.53–0.91, <0.008), the GFR rate that decreased by<60 mL/min/1.73 m was significantly lower than that of atorvastatin. Regarding long-term statin intake, GFR changed with the type of statin. This work is the first in Korea to compare each statin in terms of changes in the GFR after the statin prescription.

Application of ultra-sonication, acid precipitation and membrane filtration for co-recovery of protein

摘要: A novel method was applied to co-recover proteins and humic acid from the dewatered sewage sludge for liquid fertilizer and animal feed. The proteins in sewage sludge were first extracted using the processes of ultra-sonication and acid precipitation, and then the humic acid was recovered via membrane filtration. The extraction efficiency was 125.9 mg humic acid?g VSS volatile suspended solids (VSS) and 123.9 mg proteins?g VSS at the optimal ultrasonic density of 1.5 W?mL . FT-IR spectrum results indicated that the recovered proteins and humic acid showed similar chemical characteristic to the natural proteins and humic acid. The acidic solution (pH 2) could be recycled and used more than 10 times during the co-recovery processes. In addition, the dewatered sludge could be easily biodegraded when the humic acid and proteins are extracted, which was essential for further utilization. These findings are of great significance for recovering valuable nutrient from sewage sludge.

Numerical study of internal flow field and flow passage improvement of an inlet particle separator

摘要: By performing gas flow field numerical simulations for several inlet Reynolds numbers (from 2 × 10 to 9 × 10 ) and byflow ratios (from 10% to 20%), the present study has proposed to improve the flow passage of an inlet particle separator. An adjacent objective of the study is to lower pressure losses of the inlet particle separator (IPS). No particle has been included in the gas flow for a -epsilon turbulence model. The velocity distribution in different sections and the pressure coefficient along the duct have been analyzed, which indicates that there exist important low-velocity regions and vortices in the separation area. Therefore, the profile of streamlines along the original passage has been considered. This profile illustrated a vacuum region in the same area. All investigations suggest that the separation area is the most critical one for fulfilling the objective on pressure losses limitation. Then the flow passage improvement method has focused on the separation area. An improved shape has been designed in order to suit smoothly to the streamlines in this region. Similar numerical studies as those for the original shape have been conducted on this improved shape, confirming some considerable enhancements compared with the original shape. The significant vortices which appear in the original shape reduce in amount and size. Besides, pressure losses are greatly decreased in both outlets (up to 30% for high Reynolds number) and the flow is uniform at the main outlet. Subsequent engineering surveys could rely on expressions obtained for in both outlets which extend the pressure losses for a wide range of inlet Reynolds numbers. As a result, the numerical calculations demonstrate that the flow passage improvement method applied in this study has succeeded in designing a shape which enhances the flow behavior.
